Annex 15 Corrigendum: Small Text, Big Consequences for Cleaning Teams

EMA and PIC/S reissued the January 2026 Annex 15 concept paper in July 2026 with a new reference number and a substantially longer timetable. Nothing about cleaning validation itself changed — but the dates most teams are planning against did.

What happened

On 16 July 2026, PIC/S published a corrigendum to the concept paper on the revision of Annex 15 (Qualification and validation) of the EU–PIC/S GMP Guide: the concept paper "was submitted to a joint EMA - PIC/S public consultation and comments have been collected until 9 April 2026. Following the public consultation, the initial timetable has been revised, and only this section has been updated accordingly."

On the EMA side, the reissued concept paper now carries the reference EMA/INS/GMP/163466/2026 and still shows its original header date of 19 January 2026. The February 2026 version — preserved in the Internet Archive — carried a different reference, EMA/INS/GMP/20217/2026, and ran to six pages; the reissue runs to five. The reissued PDF contains no visible corrigendum note; the July event exists in the PIC/S announcement, not in the paper's body text.

Comparing the two texts, the substantive proposal is unchanged. What moved is the schedule:

| Milestone | February 2026 version | July 2026 reissue | | --- | --- | --- | | Consultation on the draft guideline | April 2026 | December 2026 | | Deadline for comments on the draft guideline | June 2026 | March 2027 | | Endorsement by the EMA GMP/GDP IWG | July 2026 | November 2027 | | Publication by the European Commission | by December 2026 | April 2028 | | Adoption by the PIC/S Sub-committee on GMDP Harmonisation | by December 2026 | April 2028 |

Everything else is editorial: the new reference number, page layout, and wording changes such as "the quality of AS" becoming "the quality of active substances". ECA (GMP Compliance) reported the corrigendum as dated 15 July 2026, describing minor editorial amendments, a new reference number, and a transitional period of at least six months expected to follow publication.

Why it matters

The cleaning-relevant content is untouched. The paper still proposes to extend Annex 15's scope to chemical and biological active substance manufacturers, to extend the Validation Master File, qualification-and-validation policy and change control concepts, to extend validation expectations for third-party contractors, and to extend the requirement to investigate results failing pre-defined acceptance criteria. Its stakeholder questions name cleaning validation explicitly, alongside process validation, transport validation, investigations, qualification and change control.

What changed is when any of it becomes binding. The draft guideline consultation now starts in December 2026 and the earliest credible publication by the European Commission is April 2028 — roughly sixteen months later than the original plan. For a cleaning programme that is not a regulatory change; it is a planning change. Capital requests, cleaning-remediation projects and supplier roadmaps dated against a December 2026 publication no longer sit on that clock. Teams that compressed a remediation sprint into 2026–2027 have more room; teams tempted to treat the delay as permission to stop work are wrong — the current Annex 15 applies to them today, unchanged and in force.

What is particularly interesting

Three details are worth a quality manager's minute. First, this is a corrigendum that is essentially a schedule reset: a consultation that drew substantive industry input ended with the whole programme pushed out by more than a year.

Second, a documentation trap. Anyone who cited this concept paper in a validation master file, an inspection pack or a supplier communication needs to know the reference has changed to EMA/INS/GMP/163466/2026; references still quoting EMA/INS/GMP/20217/2026 point at a superseded version.

Third, a small internal inconsistency: the revised timetable lists the concept-paper comment deadline as March 2026, while the same document's front table records 9 April 2026 — when the consultation actually closed.

What could we learn

Treat regulator reissues as change control. Before anything in a validation file, a training slide or an SOP appendix cites a guideline document, its reference number and version should have an owner, a controlled copy and a review trigger. This corrigendum is a cheap rehearsal for that habit.

Do not build strategy on consultation-stage timetables: they are forecasts, and this one moved by sixteen months after a single comment period. Separate direction from date — mandatory Annex 15 scope for API manufacturers, explicit ICH Q9(R1) integration and lifecycle expectations that cleaning validation sits inside were not weakened. Only the calendar was.

Questions for our programme

  • If Annex 15 becomes mandatory for our active substance suppliers, can we show that their cleaning validation and hold-time justification meet the standard we apply in-house?
  • Do our controlled copies of regulatory references record the correct document number — and would we notice if a text were reissued under a new reference?
  • Which of our current commitments (CAPEX, cleaning remediation, validation master plan updates) were timed against a December 2026 Annex 15 publication, and do those dates still hold?
  • How do we keep outsourced cleaning validation under our control if third-party validation expectations are formally extended?
  • Have we assessed the PIC/S PI 006-4 expectations, in force from 1 October 2026, independently of the Annex 15 timetable?

Wider context

The revision traces back to the June 2020 report Lessons learnt from presence of N-nitrosamine impurities in sartan medicines, whose chapter 4 recommends making Annex 15 mandatory for active substance manufacturers; the 115th GMDP IWG meeting (September 2024) agreed a targeted revision and alignment with ICH Q9(R1), with a comprehensive review to follow. Two weeks after the corrigendum, PIC/S published the revised Recommendations on Qualification and Validation (PI 006-4, Geneva, 30 July 2026), in force from 1 October 2026 and superseding PI 006-3 — which included the PIC/S Cleaning Validation recommendations. For cleaning teams, that is the closer document.

Validation takeaway

The July 2026 Annex 15 corrigendum changes the calendar, not the cleaning requirements. The proposal to make Annex 15 applicable to chemical and biological active substance manufacturers, and to align it with ICH Q9(R1), is unchanged; the consultation on the draft guideline now starts December 2026 and publication by the European Commission is planned for April 2028 instead of December 2026. Read it as a planning correction: refresh any controlled reference that still cites EMA/INS/GMP/20217/2026, re-date remediation and capital plans built on the old schedule, and keep working to the Annex 15 that is in force today.

Questions worth asking in our own programme

  • Have we assessed the PIC/S PI 006-4 expectations, in force from 1 October 2026, independently of the Annex 15 timetable?
